Large-Scale Testing of Bioactive Coatings for Titanium Implants by Russian Researchers

TOMSK, RUSSIA / RankWire.AI / – Russian scientists have evaluated a bioactive coating aimed at enhancing the interaction between titanium orthopedic implants and bone tissue. This material incorporates calcium phosphate sourced from hydroxyapatite and includes nitrogen compounds linked to nitric oxide synthesis. In laboratory experiments, human mesenchymal stem cells demonstrated markedly improved survival rates on coated surfaces compared to uncoated titanium. The study examined the coating’s structure, chemistry, mechanical properties, and biological response. Their peer-reviewed results were published in Applied Surface Science in 2026.

Russian team tests bioactive coating for titanium implants
Russian researchers are testing bioactive coatings designed for titanium orthopaedic implants.

Researchers at Tomsk Polytechnic University created the experimental coatings using reactive magnetron sputtering of a hydroxyapatite target inside a vacuum chamber. They varied the nitrogen-to-argon ratio during deposition to observe how each mixture influenced the surface properties. Five different conditions were tested, ranging from pure nitrogen to pure argon. The team measured parameters such as coating thickness, surface morphology, hardness, wettability, and chemical composition. Laboratory tests also assessed how live human cells responded to the modified titanium surfaces.

The experiments indicated that the argon content significantly impacted several physical characteristics of the coatings. Surfaces produced in pure argon were denser and harder than those created in pure nitrogen. As the proportion of argon increased, so did the coating thickness. Chemical analyses identified nitrogen-carbon and nitrogen-oxygen bonds present on the modified surfaces. The team then compared human mesenchymal stem cells grown on coated titanium with those on uncoated titanium, analyzing cell viability and markers related to osteogenic differentiation.

Coating evaluations reveal enhanced cell viability

The biological tests demonstrated that cell survival was notably higher on coated titanium than on uncoated samples, according to the findings. After a week, coatings with elevated nitrogen levels also suppressed activity in certain genes associated with early stages of bone-cell differentiation. Despite this, the cells maintained their ability to produce bone tissue. The research was conducted under controlled laboratory conditions using human mesenchymal stem cells, and no clinical trials or patient-based assessments of the coating’s performance have been conducted at this stage.

The biomedical evaluation was carried out by Immanuel Kant Baltic Federal University and Siberian State Medical University, with additional input from Saint Petersburg State University. The project was funded through Russia’s national science program. The researchers aimed to identify gas mixtures capable of producing coatings with optimal physical, chemical, and biological properties. Hydroxyapatite is already used in implant coatings due to its calcium phosphate composition, which closely resembles the mineral component found in human bone.

The study remains within the laboratory testing phase

The team has outlined further testing beyond the initial seven-day cell viability assessment. They plan to examine stem cell responses over 10 to 28 days and evaluate how rapidly the coatings dissolve. Additionally, they intend to measure nitric oxide release into surrounding tissues in live organisms. These future studies are not part of the current published results, which focus solely on coated titanium substrates, material properties, and in vitro biological responses, without any clinical or patient-based data.

The findings provide comprehensive laboratory insights into how the ratios of nitrogen and argon influence calcium phosphate coatings on titanium. Variations in thickness, density, hardness, chemical bonding, and cellular reactions across different gas mixtures were documented. The research confirms that coated samples support higher stem-cell survival compared to bare titanium under the tested conditions. However, as a preclinical study, these results do not establish safety or effectiveness in humans, and further biological testing will be necessary to explore properties not addressed here.
